    Eigensystem realization algorithm modal identification experiences with mini-mast

    This paper summarizes work performed under a collaborative research effort between the National Aeronautics and Space Administration (NASA) and the German Aerospace Research Establishment (DLR, Deutsche Forschungsanstalt fur Luft- und Raumfahrt). The objective is to develop and demonstrate system identification technology for future large space structures. Recent experiences using the Eigensystem Realization Algorithm (ERA), for modal identification of Mini-Mast, are reported. Mini-Mast is a 20 m long deployable space truss used for structural dynamics and active vibration-control research at the Langley Research Center. A comprehensive analysis of 306 frequency response functions (3 excitation forces and 102 displacement responses) was performed. Emphasis is placed on two topics of current research: (1) gaining an improved understanding of ERA performance characteristics (theory vs. practice); and (2) developing reliable techniques to improve identification results for complex experimental data. Because of nonlinearities and numerous local modes, modal identification of Mini-Mast proved to be surprisingly difficult.     The use of group testing to identify individuals with targeted outcomes in a population can greatly improve the efficiency, speed, and cost effectiveness of testing a population for an outcome, or at least for identifying the prevalence of an outcome in a population. The implementation of causal inference techniques can provide the basis for an observational study that would allow an investigator to gather estimates for treatment effectiveness if group testing was conducted on the population in a certain way. This thesis examines a simulation of the above outlined principles in order to demonstrate a potential application for determining treatment efficacy from observational data obtained via testing for disease outcome in a partially treated population. It is made evident that it is reasonable to make conclusions about treatment conditional incidence of an outcome in a sample of tested individuals based on outcome tests conducted on groups of individuals. Examining group study observations in the manner described in this thesis will allow researchers to estimate treatment effectiveness from partial data in situations where outcome testing may have been limited or where quick results are required from limited data

    Usability and feasibility of consumer-facing technology to reduce unsafe medication use by older adults

    Background Mobile health technology can improve medication safety for older adults, for instance, by educating patients about the risks associated with anticholinergic medication use. Objective This study's objective was to test the usability and feasibility of Brain Buddy, a consumer-facing mobile health technology designed to inform and empower older adults to consider the risks and benefits of anticholinergics. Methods Twenty-three primary care patients aged ≥60 years and using anticholinergic medications participated in summative, task-based usability testing of Brain Buddy. Self-report usability was assessed by the System Usability Scale and performance-based usability data were collected for each task through observation. A subset of 17 participants contributed data on feasibility, assessed by self-reported attitudes (feeling informed) and behaviors (speaking to a physician), with confirmation following a physician visit. Results Overall usability was acceptable or better, with 100% of participants completing each Brain Buddy task and a mean System Usability Scale score of 78.8, corresponding to “Good” to “Excellent” usability. Observed usability issues included higher rates of errors, hesitations, and need for assistance on three tasks, particularly those requiring data entry. Among participants contributing to feasibility data, 100% felt better informed after using Brain Buddy and 94% planned to speak to their physician about their anticholinergic related risk. On follow-up, 82% reported having spoken to their physician, a rate independently confirmed by physicians. Conclusion Consumer-facing technology can be a low-cost, scalable intervention to improve older adults’ medication safety, by informing and empowering patients. User-centered design and evaluation with demographically heterogeneous clinical samples uncovers correctable usability issues and confirms the value of interventions targeting consumers as agents in shared decision making and behavior change

    Adherence and Tolerability of Alzheimer's Disease Medications: A Pragmatic Randomized Trial

    BACKGROUND/OBJECTIVES: Post-marketing comparative trials describe medication use patterns in diverse, real-world populations. Our objective was to determine if differences in rates of adherence and tolerability exist among new users to acetylcholinesterase inhibitors (AChEI's). DESIGN: Pragmatic randomized, open label comparative trial of AChEI's currently available in the United States. SETTING: Four memory care practices within four healthcare systems in the greater Indianapolis area. PARTICIPANTS: Eligibility criteria included older adults with a diagnosis of possible or probable Alzheimer's disease (AD) who were initiating treatment with an AChEI. Participants were required to have a caregiver to complete assessments, access to a telephone, and be able to understand English. Exclusion criteria consisted of a prior severe adverse event from AChEIs. INTERVENTION: Participants were randomized to one of three AChEIs in a 1:1:1 ratio and followed for 18 weeks. MEASUREMENTS: Caregiver-reported adherence, defined as taking or not taking study medication, and caregiver-reported adverse events, defined as the presence of an adverse event. RESULTS: 196 participants were included with 74.0% female, 30.6% African Americans, and 72.9% who completed at least twelfth grade. Discontinuation rates after 18 weeks were 38.8% for donepezil, 53.0% for galantamine, and 58.7% for rivastigmine (P = .063) in the intent to treat analysis. Adverse events and cost explained 73.1% and 25.4% of discontinuation. No participants discontinued donepezil due to cost. Adverse events were reported by 81.2% of all participants; no between-group differences in total adverse events were statistically significant. CONCLUSIONS: This pragmatic comparative trial showed high rates of adverse events and cost-related non-adherence with AChEIs.     Ground-Based Coronagraphy with High Order Adaptive Optics

    We summarize the theory of coronagraphic optics, and identify a dimensionless fine-tuning parameter, F, which we use to describe the Lyot stop size in the natural units of the coronagraphic optical train and the observing wavelength. We then present simulations of coronagraphs matched to adaptive optics (AO) systems on the Calypso 1.2m, Palomar Hale 5m and Gemini 8m telescopes under various atmospheric conditions, and identify useful parameter ranges for AO coronagraphy on these telescopes. Our simulations employ a tapered, high-pass filter in spatial frequency space to mimic the action of adaptive wavefront correction. We test the validity of this representation of AO correction by comparing our simulations with recent K-band data from the 241-channel Palomar Hale AO system and its dedicated PHARO science camera in coronagraphic mode.Comment: To appear in ApJ, May 2001 (28 pages, 10 figs

    Mode of Action of the Natural Product Allicin in a Plant Model:Influence on the Cytoskeleton and Subsequent Shift in Auxin Localization

    Allicin is a defense substance produced by garlic cells when they are injured. It is a redox-active thiosulfinate showing redox-activity with a broad range of dose-dependent antimicrobial and biocidal activity. It is known that allicin efficiently oxidizes thiol-groups, and it has been described as a redox toxin because it alters the redox homeostasis in cells and triggers oxidative stress responses. Allicin can therefore be used as a model substance to investigate the action of thiol-specific prooxidants. In order to learn more about the effect of allicin on plants, we used pure synthetized allicin, and studied the influence of allicin on organelle movement in Tradescantia fluminensis as a cytoskeleton-dependent process. Furthermore, we investigated cytoplasmic streaming in sterile filaments of Tradescantia fluminensis, organelle movement using transgenic Arabidopsis with organelle-specifics GFP-tags, and effects on actin and tubulin in the cytoskeleton using GFP-tagged lines. Tubulin and actin were visualized by GFP-tagging in transgenic lines of Arabidopsis thaliana to visualize the influence of allicin on the cytoskeleton. Since auxin transport depends on recycling and turnover of the PIN protein involving cytoskeletal transport to and from the membrane localization sites, auxin distribution in roots was investigated using of transgenic PIN1–GFP, PIN3–GFP, DR5–GFP and DII–VENUS Arabidopsis reporter lines. Allicin inhibited cytoplasmic streaming in T. fluminensis, organelle movement of peroxi-somesperoxisomes, and the Golgi apparatus in a concentration-dependent manner. It also destroyed the correct root tip distribution of auxin, which probably contributed to the observed inhibition of root growth. These observations of the disruption of cytoskeleton-dependent transport processes in plant cells add a new facet to the mechanism of action of allicin as a redox toxin in cells

    Antidepressant Use in the Elderly Is Associated With an Increased Risk of Dementia

    A retrospective cohort study was conducted including 3688 patients age 60 years or older without dementia enrolled in a depression screening study in primary care clinics. Information on antidepressant use and incident dementia during follow-up was retrieved from electronic medical records. The Cox proportional hazard models were used to compare the risk for incident dementia among 5 participant groups: selective serotonin re-uptake inhibitors (SSRI) only, non-SSRI only (non-SSRI), mixed group of SSRI and non-SSRI, not on antidepressants but depressed, and not on antidepressants and not depressed. SSRI and non-SSRI users had significantly higher dementia risk than the nondepressed nonusers (hazard ratio [HR]=1.83, P=0.0025 for SSRI users and HR=1.50, P=0.004 for non-SSRI users). In addition, SSRIs users had significantly higher dementia risk than non-users with severe depression (HR=2.26, P=0.0005). Future research is needed to confirm our results in other populations and to explore potential mechanism underlying the observed association

    Research, Development and Education Priorities for the Aquaculture Sector in Maine

    The research priorities survey has been an ongoing initiative coordinated by the Maine Aquaculture Innovation Center in collaboration with Maine Aquaculture Association, Maine Sea Grant, and the University of Maine’s Aquaculture Research Institute since 2012. This report is based on the research priorities survey conducted in June 2019 of Maine’s aquaculture community, and is compared with surveys from 2012 and 2016 to look at trends. There have been some changes in the prioritization of needs since 2016, but many of the same research, development, and education (R&D&E) needs persist and remain unaddressed. As expected, there are different R&D&E priorities for different aquaculture sectors
